Pardon my total ignorance but after installing my new intake is it realistic to expect an additional, noticeable performance increase with a tune?

Yes, the butt dyno should feel it.

Here's something that may give you an idea, it's a spectre intake, dynomax catback, & tune, vs stock, pay close attention to the average gains in relation to the overall powerband:
http://www.expeditionforum.com/f44/couple-dyno-charts-may-help-some-folks-18573/

HP increase total: 24 rwhp (170 to 194)
TQ increase total: 26 rwtq (232 to 258)

No Tune: Avg HP: 153.8 Avg TQ: 212.7
W/ Tune: Avg HP: 167.8 Avg TQ: 231.3

You should see a similar increase assuming you have a 5.4, & the tune is similar. We dyno'd with just cai/tune vs cai/tune/catback and the catback really didn't make much difference (i dont have the chart for it, but it wasn't hardly anything.) The muffler was straight through instead of chambered though, that may have hurt it.

If whatever tuner/tuning you go with adjusts the transmission shift pressure & throttle sensitivity, you'll definitely see a difference there. You won't regret the tuning, or the JLT choice.
