https://tiresize.com/calculator/ to compare the size differences in the tires themselves, and note the changes in rpm's and speedometer readings, which will be minimal since those 2 sizes are so close....
As for how the 2 sizes will fit, that depends on the year/model of your Expy, but generally, you can add bigger tires, but only to a certain point.....and a lift may give you a little more flexibility, but the 2 main things to consider are:
A) Will the front tires rub on your fender walls and/or other suspension parts, especially at full turn, and
B) And as you said, it there enough travel when hitting bumps, rough spots, pot holes ect WITHOUT touching the insides of the fenders & slicing the tire open...
Just be aware that add a lift AND bigger tires fundamentally alters the entire geometry of the vehicle from what it was designed with, and this could be ok, or not so much.
Now, I am certainly no expert on suspension/tire changes, but there are others here who have done extensive modifications to their Expy's...some came out well, others had to do a retake and make some more adjustments, mainly because they went too far in one respect or the other
