2000 Eddie Bauer with multiple lock issues

This vehicle came with pre-existing door lock conditions, and I'm not sure where to begin the repair/ diagnoses. The Driver's side key does not work in the lock. None of the locks open consistently; using the fob, the door key, or the unlock switch on the inside of the door. It sounds like electronically, things are being communicated to each door lock, but mechanically they are all not functioning fully. The keypad on the outside also makes some transfer-type sound, but no doors open.
Gem module was tested (OK).

Where do I begin?

Based on your description, faulty door lock actuators are a likely suspect. Start by removing the panel from one of the doors that doesn't work(not the driver's because you'll probably need to go to the parts store) so you can see the lock mechanism. If you're not familiar with removing trim pieces, watch this video first because it's easy to break the various plastic tabs and connectors on the inside of the panel.

Thank you for this. It would make sense that each one of the four doors could have actuators that aren't (always) doing the job. But what about the rear hatch? The reason I ask is that when I hit 'UNlock', any, or none of the 5 doors may unlock.
I'll have a look art the video on panel removal, and then have a look at how to replace the actuator.

But what about the rear hatch?
It also has an actuator but it's a more complex assembly that controls both the liftgate and the window locks. Many problems with the rear hatch are caused not by the actuator but by the cables running between it and the locks. I'd leave that one for last.

My doors were like that, and I got some silicone spray lubricant, and sprayed the heck out of them.. They freed up and been working fine so far.. Push the easy button first

You have to remove the door panel to be able to see what's going wrong and hopefully it's not the actuator itself because replacing one is no trivial task. Some simpler causes I've encountered:
- lock handle rod bent/broken/detached
- retainer clip for the lock handle rod damaged
- E-clip for the keylock broken
- end-pieces deteriorated on the cables used by the liftgate locks
