Will I need to drain out my radiator & majority of the coolant in order to put on new heater valve? Also has anyone had issues with these leaking & pooling over the left bank or 3rd injector?

That is the top of your coolant system and you don't have to drain anything. Some fluid will come out of the heater hose (that is going to come out whether you drain or not) but you can limit that by propping the hose up while you work. I suggest you put plastic down over the engine (under the heater hoses) before starting.

And yes it is a known problem with them leaking on the 3rd and 4th cylinder area.

There is a couple different ways to burp the air back out after you have fixed it. You can look them up on you tube.
